Leeds continued their fine start to the season with a win over Tranmere at the weekend and Grayson is keen to keep the momentum going.
"We'll assess the situation but people won't want to be left out. We have to get the right balance between keeping things right," Grayson told the club's official website.
"If everyone is fit and available then we will go with what we have. The players are thriving and playing with confidence."
Grayson will once again be without left-back Ben Parker, who has missed the last four games with a hamstring injury and centre-back Richard Naylor, who has been sidelined since undergoing a back operation at the end of last season.
Watford look set to be without Jobi McAnuff and Nathan Ellington while there is new speculation concerning Tommy Smith ahead of the trip to Leeds.
Winger McAnuff has a back injury and striker Ellington a knee problem, and neither are likely to travel having missed Saturday's 1-1 draw with Blackpool.
Hornets fans may also have seen the last of Smith, with Reading expected to resurrect their £1.8million move for the striker.
Midfielder Don Cowie suffered no ill-effects following his return from a hamstring injury at the weekend and he will be involved once more.
Loan duo Henri Lansbury and Tom Cleverley have been given permission to play in the competition by parent clubs Arsenal and Manchester United.
